Sirtris Pharmaceuticals: Resveratrol reduces fat and boosts endurance by activation of SIRT1, first aging gene shown to accelerate human metabolism

Findings published in Cell suggest broad implications for the treatment of diseases of aging such as diabetes

CAMBRIDGE, MA and Strasbourg, France ¨C November 16th, 2006 ¨C Sirtris Pharmaceuticals and the University Louis Pasteur, Strasbourg announced that in an article published today in Cell, ¡°Resveratrol improves mitochondrial function and protects against metabolic disease by activating SIRT1 and PGC-1¦Á.¡± Lagouge et al., Cell. 2006; 127: 1¨C14, SIRT1 was shown for the first time in a human population to accelerate metabolic rate.

In a human population in Finland, SIRT1 was linked to increased energy expenditure as demonstrated by genetic studies of three variants of the SIRT1 gene. The study also showed that treating mice with resveratrol increased mitochondrial biogenesis leading to increased exercise endurance and protection from diet induced obesity. Activation of SIRT1, the best characterized of the recently-discovered family of sirtuin enzymes, was shown to be the mechanism by which these therapeutic benefits occur. Mice were dosed with 200 mg/kg or 400 mg/kg of resveratrol daily in either normal chow or high fat chow. The mice on resveratrol lost weight due to decreased fat, and this was attributed to an increase in the number and function of mitochondria. The resveratrol-treated mice also exhibited improved insulin sensitivity and an increased metabolic rate. Notably, mice treated with resveratrol showed a two-times increase in exercise endurance. These effects were shown to be mediated through SIRT1 and PGC-1¦Á.

The authors of the Cell article include the teams of the principal investigator Johan Auwerx, M.D. Ph.D., Professor at the Medical Faculty in Strasbourg, at IGBMC (Unit¨¦ mixte de recherche CNRS, Inserm, University Louis Pasteur), France, and of Pere Puigserver, Ph.D. from Johns Hopkins University School of Medicine in Baltimore (now at the Dana-Farber Cancer Institute/Harvard Medical School in Boston), both members of the Scientific Advisory Board of Sirtris Pharmaceuticals, and Sirtris scientists: Peter Elliott, Ph.D. Senior Vice President and Head of Development, Phil Lambert, Ph.D. Senior Director of Pharmacology, and Jill Milne, Ph.D., Senior Director of Biology.

"This work is significant because it shows that a SIRT1 activator can protect against metabolic disease, highlighting the therapeutic potential of sirtuins. Resveratrol a compound found in the skin of red grapes and hence in red wine, could very well explain the French Paradox," said Johan Auwerx.

Sirtris has initiated a human Phase 1b clinical trial in diabetes with SRT501, a proprietary formulation of resveratrol with improved bioavailability. SRT501 is the first small molecule to enter human clinical trials that is designed to activate SIRT1. Sirtris has applied this scientific discovery to the development of SRT501, which activates SIRT1, for the treatment of diseases of aging such as metabolic and mitochondrial disorders. In addition, Sirtris has a robust pipeline of novel small molecule drug candidates that are potent SIRT1 activators and are chemically distinct from resveratrol.

The 50 Day Blink 182 Tour Schedule Begins Selling Tickets on May 30, 2009


The band Blink-182 will be coming back for an anticipated journeying during the 2009 summer. The Blink-182 journeying schedule will all get underway on Friday, July 24th, and culminate in an October 3rd show at the Borgata Events Center in Atlantic City, New Jersey. Along the way the Blink-182 tour

dates will include 50 different show dates, and will take place at various venues across the country, including a few shows within Las Vegas. The journeying has been a daylong time coming, and now the group will re-unite for a daylong extension during the summer concert season.

The Blin-182 journeying is produced by Live Nation, and won't just be the group that is performing at all of the events. Other artists that will be joining them at given stops along the way include Weezer, Fall Out Boy, Panic at the Disco, All-American Rejects, Taking Back Sunday, Asher Roth, and Chester French just to name a few. Some of the supporting groups may even be bigger draws than Blink-182 themselves, especially the All American Rejects that have been making their presence known on Top 40 radio around the country recently.

After Travis Barker was in a plane crash last year, it was thought that they wouldn't be doing a journeying this year, but it seems that Barker is doing better, and that the group had the urge to get back on the road. Members Mark Hoppus, Travis Barker, and Tom DeLonge will all be on hand for the trek across the country, and it is sure to highlight some of the best music Blink-182 has to offer. It has been five years since they toured together, so this will be a thin instance for fans to get a glimpse of the band, and it will be a good stop-gap until the band releases a new album that has been rumored to be in the works for some time now.

Tickets for the upcoming Blink-182 journeying dates go on sale May 30th on the Live Nation web-site (livenation.com) and some of them are thought to be quite affordable at around $20.00 for a couple of the dates. This will be a chance for fans of the group to see the group again, and see if the music is as strong as they once believed. It will also be a treat to see some of the great acts that will join them this summer.

David Cooks Brother

David cooks brother, Adam Cook after fighting cancer for more than a decade, David Cook's brother, Adam, died Saturday, the American Idol winner announced Sunday morning before the 12th annual Race for Hope 5K in Washington, D.C.

"I actually lost my brother yesterday to a brain tumor and I couldn't imagine being anywhere else right now," Cook, 26, told the crowd at the benefit for brain cancer research (see video here). Cook was both serving as grand marshal and running with a team that raised $98,000.

Cook's brother, Adam, 37, had battled the disease throughout Cook's American Idol competition, and had the chance to see his younger sibling perform in April 2008 on the famous stage. At the time, Adam, a married father and lawyer in Terre Haute, Ind., was undergoing chemotherapy treatment.

"I'm not going to lie to you. He still has a tough road ahead of him," Adam's longtime friend Darrick Scott had told PEOPLE. "But this trip to Los Angeles has put a whole new pep in his step."

The Idol judges had even noted that having Adam in the front row may have given David the inspiration needed to win.

Simon Cowell on Sunday offered his condolences. “I was incredibly saddened to hear the news," he said in a statement to PEOPLE. "I met Adam Cook a few times at American Idol. He was an incredible guy, and right now my thoughts are with David and his family.”

In March this year, the Idol champ Cook canceled two shows on his current tour "to deal with personal family matters." His rep had told PEOPLE that he had recently been spending time with his brother.

For Sunday's race, the singer flew into the capitol after a performance Saturday night at Sun Fest in Palm Beach, Fla., where he had spent most of the day hanging out. His plan was to take a 3 a.m. flight.

"I don't run normally, but it seems a small sacrifice compared to what those who have fought cancer have endured," Cook earlier had told PEOPLE. "And the fact that my fans have rallied to raise so much in donations is the biggest compliment."

At the early-morning benefit, he said, "I lost one today," addressing all the runners, supporters and organizers, "but I gained 9,000 [friends] and I will be here every year that they will have me."

Adam Cook

Adam Cook


Adam Cook is the brother of American Idol winner David Cook. The 36-year-old Cook battled brain cancer, which reportedly spread to his spine. He had been undergoing chemotherapy treatments.

Adam cook passed away at 36 years old in the early hours of May 3, 2009. His death was announced by his brother David Cook while opening the Race for Hope in Washington D.C., a benefit race for brain cancer.

Adam Cook was first diagnosed with brain cancer in 1998. He is currently battling his second diagnosis of cancer which has spread to his spine and brain.

Cook was a lawyer up until his condition worsened. Cook was sick in bed before the news that he would be able to see his brother play.

Despite his illness, Adam Cook was able to attend the Idol finale on May 21, 2008 to see his brother win the competition. He and wife Kendra Cook flew to Los Angeles from their home in Terre Haute, Indiana, on a private plane. Reports say the flight was paid for by an anonymous private donation.

Adam Cook was able to see his brother compete twice during the season. In April of 2008, Adam Cook flew to California on a chartered medical jet with medical staff handy. The flight was paid for by his friends and a local councilman. A local radio station provided a paid hotel stay across the street from the studio where Idol tapes.
